چکیده

The intracellular concentration of trehalose increases in the stationary-phase cells of Escherichia coli. The effects of trehalose on transcription in vitro by E. coli RNA polymerase were compared for two holoenzymes, Es and Es, which were reconstituted from purified core enzyme and either s (the major s at the exponential growth phase) or s (the essential s at the stationary growth phase), respectively. The optimum trehalose concentration giving maximum transcription by Es was higher than that by Es. Transcription activation by trehalose was attributed to both increased formation of Es holoenzyme and increased transcription initiation by Es from s-dependent promoters. The activation of Es by trehalose was additive with the transcription enhancement by decreased superhelicity of template DNA prepared from stationaryphase cells. We thus propose that the selective activation of transcription by Es holoenzyme takes place in the presence of specific conditions and factors present under stress conditions.

Oligomerization of the E. coli Core RNA Polymerase: Formation of (α2ββ'ω)2–DNA Complexes and Regulation of the Oligomerization by Auxiliary Subunits

In this work, using multiple, dissimilar physico-chemical techniques, we demonstrate that the Escherichia coli RNA polymerase core enzyme obtained through a classic purification procedure forms stable (α(2)ββ'ω)(2) complexes in the presence or absence of short DNA probes. Multiple control experiments indicate that this self-association is unlikely to be mediated by RNA polymerase-associated non...

MINIREVIEW / MINI-SYNTHÈSE Regulation in the rpoS regulon of Escherichia coli

In Escherichia coli, the transcription factor σS, encoded by rpoS, controls the expression of a large number of genes involved in cellular responses to a diverse number of stresses, including starvation, osmotic stress, acid shock, cold shock, heat shock, oxidative DNA damage, and transition to stationary phase. A list of over 50 genes under the control of rpoS has been compiled. The transcript...

Promoter Selectivity of Escherichia coli RNA Polymerase s Holoenzyme Involved in Transcription of Flagellar and Chemotaxis Genes

The rpoF gene of Escherichia coli codes for the RNA polymerase s (or s) subunit, which is involved in transcription of the flagellar and chemotaxis genes. Both s and s (the major s subunit in growing cells) were overexpressed, purified to homogeneity, and compared with respect to activity and specificity. The affinity of s to core RNA polymerase (E) is higher than that of s, as measured by gel ...

The reversible denaturation of deoxyribonucleic acid-dependent ribonucleic acid polymerase of Escherichia coli.

RNA polymerase core and holoenzyme isolated from Escherichia coli have been denatured and reconstituted in high yield (70 to 100%). Ultracentrifugation studies and measurement of tryptophan fluorescence indicate that the reconstituted enzyme has a structure similar to that of the native enzyme. The reconstituted holoenzyme transcribes T7 DNA as efficiently as the native holoenzyme. RNA synthesi...

Purification of Bacillus subtilis RNA polymerase with heparin-agarose. In vitro transcription of phi 29 DNA.

We have devised a new procedure for the purification of highly active preparations of Bacillus subtilis RNA polymerase holoenzyme. A column of heparin-agarose A-15m is used to rapidly and quantitatively adsorb RNA polymerase from the initial crude extract fraction. This affinity procedure obviates the necessity of including nucleic acid precipitation or partitioning steps and allows for rapid s...
